Recombinase Aided Amplification (RAA) is an isothermal RNA and DNA amplification & detection technology developed and manufactured by Jiangsu Qitian Gene Biotechnology Co., Ltd. It provides rapid molecular results within minutes. The amplified nucleic acids can be detected in one of three ways: through real-time fluorescence detection, through analysis using lateral-flow strips, or through detection using agarose gels or capillary electrophoresis.

Which heating device should I use for nucleic acid extraction using SwiftX™ kits?

Heat incubation using direct contact heating devices is preferred. We have validated dry heat blocks and water baths as reliable heat sources. In contrast, heating ovens are not suitable for providing the heat for optimal nucleic acid extraction to your samples. We found that the conduct of heat into the sample is not sufficient, even if the oven is providing air ventilation. For further information please have a look into Application Note 2021-01, which you find on the product page of SwiftX™ Swabs.

 

Can I shorten the time of the heat incubation indicated in the instructions of SwiftX™ kits?

It depends on the actual extraction kit. With “SwiftX DNA” the duration of the heat incubation depends on your experimental setup. For “SwiftX Swabs” and “SwiftX Media” the time and temperature of heat incubation should not be shortened, because the heating step in the protocols of SwiftX Swabs and SwiftX DNA has two functions. In the first few minutes of incubation the enzymes in the lysis buffer are activated and enable an efficient degradation of inhibitors such as RNases. In a later phase of the heat incubation, the same enzymes are irreversibly inactivated themselves, so that they do not interfere with your downstream application. It is important to note that proper enzyme inactivation is dependent on your choice of the right heating device.
