Cost-effective, Rapid DNA extraction
SwiftX™ Hi-Sense is designed for sensitive and rapid extraction of DNA from bacteria and viruses as well as genomic DNA from liquid human or animal samples that contain low numbers of cells and virus particles such as saliva, urine and swabs in transport media (e.g. oral, nasal, and environmental swabs).
SwiftX™ Hi-Sense has been validated for higly sensitive detection of African Swine Fever (ASF) Virus.
Simple and streamlined rapid DNA extraction kit
Workflow for cell capture, extraction and reverse purification:
- mix sample with Buffer EN and Beads B
- incubate for 3 minutes at ambient temperature
- place tubes in a magnetic rack
- discard supernatant
- wash Beads B once with Buffer EN (could be obmitted, depending on the specimen type)
- resuspend Beads B in Buffer DL
- incubate for 5 to 15 minutes at 95°C
- remove tubes from heat block and place in a magnetic rack
- use supernatant for downstream application

Technical materials and resources

Centrifugation-free cell and virus capture
Our proprietary centrifugation-free cell capture technology is unique among nucleic acid extraction solutions available on the market. It can be utilized to concentrate human & animal cells, parasites, and bacteria in liquid samples in just 4 minutes. Our capturing technology enriches intact cells and virus particles but not free DNA & RNA. This is the reason why SwiftX Hi-Sense provides an excellent discrimination between naked DNA/RNA and DNA/RNA originating from intact cells and viruses.
Reverse Purification
One of the two differentiators to most rapid DNA extraction methods is our Reverse Purification technology, which actively removes inhibitors and cell debris from the cell lysate containing the DNA. This is why our extraction protocols works well with much more sample types than direct extraction methods of competitors. Our Reverse Purification is performed using proprietary magnetic particles and does not need bulky or expensive equipment such as a centrifuge or addition of Chelex resin - a simple magnetic stand is sufficient.

Frequently Asked Questions
Can I shorten the heat lysis step in SwiftX™ Hi-Sense?
The duration of the heat incubation for DNA extraction in SwiftX Hi-Sense has been set to 10 minutes to ensure that the target cells are sufficiently lysed and their DNA is released. If you are looking for even faster DNA extraction you may shorten the heat incubation to as little as 5 minutes, but you need to keep in mind that this will also lower the DNA extraction efficiency.
Does heat lysis for 10 minutes harm or degrade my DNA?
No worries, it does not. An incubation at 95°C is not intefering with the DNA integrity apart from melting the double strands. Also, Buffer DL is free of substances or conditions that are capable of degrading DNA.
What sample types can be extracted with SwiftX™ Hi-Sense?
SwiftX Hi-Sense was designed to be applied to liquid specimens, which contain a low number of cells and little amounts of proteins. Examples are oral fluid and saliva, but also urine and swabbed samples eluted in transport media. Blood serum, however, can be extracted too, but SwiftX Hi-Sense is not the optimal extraction kit for it.
What sample volumes can be processed with SwiftX™ Hi-Sense?
The standard sample volume, which can be processed by SwiftX Hi-Sense is 1mL. If you want to extract larger volumes of liquid, additional Buffer EN is needed. For extraction from e.g. 2mL oral fluid, the sample need to be mixed with 1.4mL Buffer EN and 30µL Beads B (the amount of Beads B is not increased!).
Buffer EN can be purchased as separate accessory from Xpedite Diagnostics.
Which cells or structures can be captured and enriched with SwiftX™ Hi-Sense?
Out magnetic particles Beads B enable a capture of various cells without the need for centrifugation. They bind to different cell types of human and animal origin such as epithelial cells. They also bind viral particles and bacteria, but the spectrum of species is limited. Beads B are known to bind viruses such as the African Swine Fever (ASF) Virus and Respiratory Syncytial Virus (RSV) and bacteria such as Mycoplasma, Staphylococci and Leptospira.
How does Reverse Purification works?
The process, which we named “Reverse Purification”, is a true paradigm change in nucleic acid extraction. Instead of binding nucleic acids to magnetic particles, impurities such as proteins, cell debris and others are bound to our magnetic particles Beads A and extracted DNA and RNA remains untouched in the lysate. The inversion of the conventional nucleic acid purification workflow minimizes has two significant advantages: (a) there is no loss of DNA and RNA during the extraction and purification process, which prevents reduced yields of nucleic acids, and (b) SwiftX™ DNA can be appplied to a much wider range of sample types than other rapid extraction technologies, which do not feature a reverse purification of extracted DNA or RNA.

Are Beads B of SwiftX™ Hi-Sense binding DNA or RNA?
No, Beads B are designed to capture intact cells and viral particles. They are not capable of binding nucleic acids, neither RNA nor DNA. Due to their chemical properties, they are used for clearance of the lysate after the heat extraction step by removing cell debris and other inhibitory substances.
